Peas (Pisum sativum L.) are exposed to waterlogging at germination when grown as relay in rice‐based cropping. Ninety‐one germplasm accessions were evaluated in relay (sown in waterlogged soil), and subsequently 10 diverse genotypes compared under relay and sole cropping (conventional tillage sowing) over two seasons in Bangladesh. Contrasting genotypes, BM‐3, NL‐2 and Kaspa, were further evaluated in three waterlogging treatments (drained control, 4 and 8 days waterlogging) in the glasshouse. Conspicuous variation in waterlogging tolerance at germination was observed in the field and confirmed under controlled conditions. In relay sowing in 2011, emergence of a few genotypes was affected by waterlogging. In 2012, emergence in relay was severely affected (12 plants/m2) compared to sole sowing (37 plants/m2). Among genotypes BM‐3 had 6 plants/m2 emerge, which all subsequently died, in contrast to NL‐2 in which emergence was 13 plants/m2 with all plants surviving. In the glasshouse, there was 14% emergence in BM‐3, 40% in NL‐2 and 55% in Kaspa after 8 days of waterlogging. Such marked differences in waterlogging tolerance at germination in the model pea are the first reported and illustrate prospects for selection to improve adaptation to relay sowing in South Asia.  相似文献

A field experiment was performed to explore responses of carbon metabolism, antioxidant system and endogenous hormone content of summer maize hybrids DengHai605 (DH605) and ZhengDan958 (ZD958) to waterlogging at the third leaf stage (V3), the sixth leaf stage (V6) and the 10th day after the tasselling stage (10VT). Results showed that waterlogging significantly decreased the contents of zeatin riboside (ZR), indole‐3‐acetic acid (IAA) and gibberellic acid (GA), compared to those of CK. However, leaf abscisic acid (ABA) content was significantly increased by waterlogging at different stages, with the most significant increase was found in the treatment of waterlogging at V3 (V3‐W), with an increase of 30% and 29% for DH605 and ZD958, respectively. Waterlogging significantly decreased antioxidative enzyme activities, accelerating leaf senescence, resulted in the disorder of leaf gas exchange parameters and chlorophyll fluorescence parameters. In addition, waterlogging decreased key enzyme activities of carbon metabolism (ribulose bisphosphate carboxylase and phosphoenolpyruvate carboxylase), with the most significant reduction in V3‐W, with a decrease of 46% and 49% for DH605, and 53% and 55% for ZD958, respectively. Visibly, waterlogging disturbed carbon metabolism, affected plant endogenous hormone content, accelerated leaf senescence and eventually resulted in a significant reduction in photosynthetic characteristic and grain yield. V3 was most susceptible to waterlogging, followed by V6 and 10VT.  相似文献

As suggested by the Office International des Epizooties (OIE), fishes belonging to the genus Oplegnathus are more sensitive to megalocytivirus infection than other fish species including red sea bream (Pagrus major). To assess the roles of the innate immune response to these different susceptibilities, we cloned the genes encoding inflammatory factors including IL‐8 and COX‐2, and the antiviral factor like Mx from red sea bream for the first time and performed phylogenetic and structural analysis. Analysed expression levels of IL‐1β, IL‐8 and COX‐2 and the antiviral factor like Mx genes performed with in vivo challenge experiment showed no difference in inflammatory gene expression or respiratory burst activity between red sea bream and rock bream (Oplegnathus fasciatus). However, the Mx gene expression levels in red sea bream were markedly higher than those in rock bream, suggesting the importance of type I interferon (IFN)‐induced proteins, particularly Mx, during megalocytivirus infection, rather than inflammation‐related genes. The in vitro challenge experiments using embryonic primary cultures derived from both fish species showed no difference in cytopathic effects (CPE), viral replication profiles, and inflammatory and Mx gene expression pattern between the two fish species.  相似文献

Cylindrospermopsin (CYN) is a cyanotoxin that is cytotoxic to a wide variety of cells, particularly to the hepatocytes. In this study, the toxic effects of purified CYN were investigated in primary cultured hepatocytes of Neotropical fish Hoplias malabaricus. After isolation, attachment, and recovery for 72 h, the cells were exposed for 72 h to 0, 0.1, 1.0, 10, and 100 μg l?1 of CYN. Then, cell viability and a set of oxidative stress biomarker responses were determined. Catalase, superoxide dismutase, glucose-6-phosphate dehydrogenase, and glutathione S-transferase activities were not affected by exposure to CYN. Concentration-dependent decrease of glutathione reductase activity occurred for most CYN-exposed groups, whereas non-protein thiol content increased only for the highest CYN concentration. Lipid peroxidation, protein carbonylation, and DNA damage levels were not altered, but reactive oxygen species levels increased in the cells exposed to the highest concentration of CYN. Cell viability decreased in all the groups exposed to CYN. Thus, CYN may cause a slight change in redox balance, but it is not the main cause of cell death in H. malabaricus hepatocytes.  相似文献

Marine fisheries target and catch fish both for direct human consumption (DHC) as well as for fishmeal and fish oil, and other products. We derived the fractions used for each for 1950–2010 by fishing country, and thus provide a factual foundation for discussions of the optimal use of fisheries resources. From 1950 to 2010, 27% (~20 million tonnes annually) of globally reconstructed marine fisheries landings were destined for uses other than DHC. Importantly, 90% of fish destined for uses other than DHC are food‐grade or prime food‐grade fish, while fish without a ready market for DHC make up a much smaller proportion. These findings have implications for how we are using fish to feed ourselves or, more appropriately, how we are not using fish to feed ourselves.  相似文献

Methionine–choline‐deficient (MCD) mammals are known to accumulate liver TAG probably due to phosphatidylcholine (PC) deficiency and thus assembly of VLDL and transport of lipids from liver to peripheral organs. To assess whether supplementation of choline could spare methionine and secure a healthy liver metabolism, by reducing the endogenous PC synthesis without interfering with lipid transport and distribution, Atlantic salmon with initial BW of 700 g were fed adequate (1.9 g Met/16 gN) or surplus methionine (2.5 g Met/16 gN) diets of which were supplemented with choline or not for a period of 19 weeks. Fish fed the low‐methionine diets had reduced growth (p = .013) due to reduced protein gain (p = .007), while lipid gain slightly improved in fish fed the choline‐supplemented diets (p = .047). Also, feed conversion improved when fed surplus methionine (p < .001), while choline supplementation had no impact on feed conversion. No interaction between choline and methionine on growth performance or retention existed. Phospholipid status in liver and muscle was not affected by treatments, and no liver TAG accumulation occurred at the methionine levels used. Gene expression of ApoB100 necessary for assembling VLDL or pemt necessary for endogenous PC synthesis was unaffected by treatments. Capacity of methylation (MAT, BHMT) within the liver was not affected by treatment nor was the gene expression of enzymes in liver transsulfuration (CBS or CDO). Methionine status within liver was unaffected by treatments, while free methionine reduced in those fish fed the low‐methionine diets in muscle and plasma. Cystathionine and taurine were elevated when fed surplus methionine. Choline supplementation had no impact on sulphur amino acid metabolites in either tissue. Neither did choline supplementation improve TAG mobilization from liver to muscle. To conclude, choline does not improve endogenous phospholipid synthesis or transport of TAG from liver to muscle depot when added to diets containing 1.9 g Met/16 gN, while surplus methionine improved growth and protein retention, indicating that 1.9 g Met/16 gN is enough to support a healthy liver metabolism, but too low to support muscle protein deposition in adult salmon fed high plant protein diets for longer periods of time.  相似文献

This study examines how appetite and growth regulation of Atlantic salmon are affected by low dissolved oxygen (LO) and dietary digestible energy levels (DE: high [HE] vs. low [LE]). Long‐term exposure to LO resulted in a reduced feed intake, growth, digestible protein and fat retention efficiencies and increased feed conversation ratio and plasma ghrelin concentrations (< .05) compared to high dissolved oxygen (HO). Pair‐feeding of rations based on the feed intake of the LO groups, but fed at HO, resulted in a 50% growth improvement in HE diet groups. This suggests that the poor growth under LO was not entirely caused by the reduced feed intake. Salmon adapted to LO by increased haemoglobin concentrations, while osmoregulation was affected by increased plasma chloride concentrations (< .05). Plasma ghrelin concentration was unaffected by DE (> .05). Growth regulation was affected by the HE diet, with increased liver and muscle growth hormone receptor ghr1 mRNA (< .05), regardless of oxygen level. The growth depression due to low oxygen appears to be related to higher metabolic costs, while higher DE upregulates the GH‐IGF system at the ghr1 level and found to be beneficial for growth, feed intake, oxyregulation and osmoregulation under hypoxia.  相似文献

The incidence of fish pathogenic oomycetes, Saprolegnia, has increased significantly in aquaculture since the ban of malachite green. For the efficient characterization of anti‐Saprolegnia therapeutics, simple accurate methods are required. However, the current screening methods are limited by time, and none of them are confirming the viability of treated spores or hyphae. In this study, a modified fluorescence‐based assay for the in vitro screening of Saprolegnia inhibitors has been developed. This method involves the use of FUN‐1 viability dye combined with calcofluor white M2R, and is based on the formation of orange‐red cylindrical intravacuolar structures (CIVS) in metabolically active spores, hyphae and biofilms. Heat‐killed and bronopol‐treated Saprolegnia spores, hyphae and biofilms exhibited diffuse bright green fluorescence which confirms complete loss of viability. For boric acid‐treated spores, no germination was observed. However, tiny CIVS were observed in 50% of treated spores which indicated reduction in their viability. Our results proved that FUN‐1 dye is an efficient tool to distinguish between live and dead Saprolegnia spores, hyphae and biofilms and to monitor the change in Saprolegnia viability during qualitative evaluation of potential anti‐Saprolegnia compounds.  相似文献
